  • Study on the Cellular Immunological Regulation of Sheep Placental Transfer Factor Injection

    [Objective] The research aimed to study the effects of sheep placental transfer factor injection on the cellular immunity of animals. [Method] Taking rabbits and Kunxi mice as test objects, the effects of sheep placental transfer factor injection on the quantity and activity of T lymphocyte in rabbit and its activation on the lymphocyte of mice were detected by using E rosette test and lymphocyte transformation test. [Result] Sheep placental transfer factor injection had the functions of significantly increasing the quantity and activity of lymphocyte, which could obviously enhance the transformation rate of lymphocytes. [Conclusion] Sheep placental transfer factor injection could improve and enhance the specific cellular immunity of test animals.
    Study on the Content of Several Kinds of Heavy Metals in the Soil with Different Crop Rotation Years of Panax notoginseng

    The content of several kinds of heavy metal elements in the soil after planting Panax notoginseng was detected and the residual situations of heavy metal elements in the soil with different crop rotation years of P. notoginseng were understood, so as to provide reference basis for planting P. notoginseng in the future. The soil cultivated with the first, third, fifth and seventh planting year of P. notoginseng were collected from the production areas of P. notoginseng. The content of Cu, Zn, Pb, Cr and Ni in the soil was detected by using flame atomic absorption method. The results showed that the content of these kinds of heavy metals in the soil was accordant with national soil standards and GAP cultivation standards of P. notoginseng. And the content of Cu, Zn, Pb, Cr and Ni in the soil with different crop rotation years didn′t show regular changes.

    The effects of cysteamine on GH, SS and IGF-1 in the serum of Tibetan Sheep weaned lamb were studied. 40 80-day-old weaned lambs of Tibetan Sheep with the average initial weight of 16 kg(half male and half female) were randomly divided into 2 groups. 300 mg/(kg·BW) of CS was added in the concentrate of diet every two days. The trial lasted 42 days. The blood samples were collected from empty stomach of sheep by jugular vein in the morning of the 0th, 7th, 14th, 21st, 28th and 35th day after medication. The content of GH, SS and IGF-1 in the plasma was determined by radioimmunoassay. The results showed that adding CS in the concentrate in the test group could increase the content of GH and IGF-1 in the serum of weaned lambs of Tibetan Sheep to different extent and there was significant difference between test group and control group(P〈0.05). SS decreased to different extent and SS content in test group had significant difference with that in control group from the 3rd week to the 42nd day.

    The production indices (dry matter production,tiller number and plant height, etc.) of grassland with different germination rate (1%,2%,5%,10%and 15%)of Setaria sphacelata cv. Narok in the second planting year were determined in 2013. The results showed that there was no significant difference of the production characteristics of grassland with different germination rate of S. sphacelata cv. Narok in the second planting year, due to its strong tillering compensation capacity.

    Mercury content in seaweed was determined by microwave digestion -hydride generation atom fluorescence spectrometry.The results showed that mercury content in seaweed was 19.46 μg/ kg and the recovery rate of mercury in the standard substance spinach was 102%-106%. The determination method was simple,rapid, with good linear correlation, lower detection limit, higher recovery and accurate determination results.

    The purpose of this study was to establish an effective method for detecting the expressed product by Escherichia coli transformed byβ-mannanase. Due to little expression amount and the changes of product structure, the conventional detection method couldn′t accurately judge the properties of expression product. So the selfmade antigen and anti-serum were used to establish ELISA. The detection results showed that E. coli transformed byβ-mannanase could successfully expressβ-mannanase.
    Nutrients of Bottom Mud in Culture Pond and Its Effects on the Productivity of Spinacia oleracea L.

    The yield of bottom bud in culture pond in Datong and Yuncheng of Shanxi Province was measured. And the contents of nutrient salt in the bottom bud and its surrounding soil were compared. Finally the productivity of Spinacia oleracea L. in bottom bud and its surrounding soil was studied. The results showed that the sediment amounts of bottom bud in the ponds of Datong and Yuncheng were 19.18 and 19.98 kg/m2 respectively. They were both higher, but there was little difference. The results of nutrient salt in the bottom mud and its surrounding soil of pond showed that the content of nutrient salt in the bottom mud of pond was obviously higher than that in the surrounding soil of pond. And the content of nutrient salts in test materials obviously decreased after planting S. oleracea. The productivity of S. oleracea in test group and control group were higher than that in the surrounding soil of pond, but there was little difference. Therefore the surrounding soil of pond could also be used for planting and the bottom mud in test group and control group could be used in the improvement of low-fertility soil.
    Determination and Analysis of Harmful Heavy Metals in Soil of Beef Cattle Farmland in Yili Region

    [Objective] The research aimed to determine the soil environmental indices in beef cattle farm in Yili region of Xinjiang and evaluate soil environmental quality conditions of beef cattle farm. [Method] Soil samples was collected from representative points in four breeding farms to determine the content of harmful substances (total Pb, total As, total Hg, total Cd and total Cr). And the soil quality of breeding farms was evaluated. [Result] The average content of total Cr, total Pb, total As, total Hg and total Cd in the soil of four breeding farms were 29.352, 34.644,0.250,0.138 and 0.550 mg/kg respectively, which were lower than the limited concentration of Farmland Environmental Quality Evaluation Standards for Livestock and Poultry Production (HJ 568-2010). [Conclusion] The soil environmental quality of four beef cattle farms was at clean level, which was accordant with the soil environmental quality standard of beef cattle breeding.
    Enzyme Kinds of Dominant Cellulose-degrading Bacteria in Camel Rumen and Detection Methods Comparison of Its Activity

    The kinds and activity of cellulase of 7 strains of dominant cellulose-degrading bacteria separated from healthy camel rumen were determined by using traditional determination method of cellulase activity 3, 5-dinitrosalicylic acid method (DNS) and self-constructed determination method of cellulase activity. The determination results showed that the activity of CX in different strains determined by DNS was 1#〉2#〉17#〉10#〉15#〉8#〉20#. And the activity of C1 was 1#〉2#〉17#〉20#〉10#〉15#〉8#. The activity ofβ-glucosidase was 1#〉2#〉17#〉10#〉20#〉8#〉15#. Total enzyme activity was 1#〉2#〉17#〉8#〉10#〉15#〉20#. It was seen from the determination results by the self-constructed method that the activity of CX in different strains was (1#、2#、15#)〉(10#、17#)〉(8#、20#). And the activity of C1 was (1#、2#、15#、17#)〉(8#、10#)〉20#. The activity of β-glucosidase was (1#、2#、8#、10#、15#、17#)〉20#. Total enzyme activity was 2#〉1#〉15#〉17#〉10#〉8#〉20#. In conclusion, the enzyme activity determined by two methods was adjacent. But there were a great number of problems in the determination process by DNS method. And the self-constructed method could conquer the disadvantages of DNS method. The determination results of self-constructed method were simple and intuitive. And the determination results would be more accurate when the self-constructed method was combined with DNS method.

    213 sheep were collected from 4 banners (counties) of Inner Mongolia to detect sheep paratuberculosis for three times by using four detection methods of AGID, DTH, CF and ELISA. The detection rate of four detection methods was compared. The results showed that the detection rate of AGID, DTH, CF and ELISA were 3.8%, 4.7%, 5.2% and 11.3% respectively. The detection rate of ELISA was the highest and this method was simple and sensitive and had actual application and popularization values. The research had some guiding significance for the detection methods selection of sheep paratuberculosis in Inner Monoglia.

    In order to study the evolution characteristics and influence factors of meteorological elements and the forming causes of snowstorms, a snowstorm disaster weather process that occurred during March 3rd-4th, 2007 was analyzed by using conventional meteorological data, satellite cloud image and numerical forecast products, etc. The results showed that the key factors that inspired such disaster weather were favorable large-scale circulation, abundant water vapor conditions, coupling of upper-lower jets, confluence of cold and warm low-level jets, strong upward movement and momentum downwash role. Some experiences and indices were summarized by analyzing the big snowfall process and it was applied in the forecast in the future, so as to provide reference basis for the forecast of rainstorm in the future.

    Taking aged female Wister rats as research objects, the effects of adding different deer fetus preparations in normal drinking on the immune indices of aged female rats were studied. The results showed that adding deer fetus preparations in the diet could significantly increase the content of total protein in the serum of aged female rats and enhance the body immunity of aged female rats. The effect of formula 2(deer fetus powder, ginseng, Codonopsis pilosula and bee pollen) was the best.
    Effects of Ethanol Extracts from Allium mongolicum on the Production Performance of Broilers

    120 one-day-old Ross 308 commercial broilers were randomly divided into 4 groups, with three repetitions in every group and 10 broilers in every repetition. The broilers in control group were fed with the basic diet and that in test group AWEⅠ, AWEⅡand AWEⅢwere fed with the diet with adding 100, 300 and 500 mg/kg ethanol extracts from Allium mongolicum. The effects on the production performance of broilers were studied. The results showed that ethanol extracts from A. mongolicum had no significant effects on the production performance of broilers (P〉0.05), but it had the functions of increasing daily weight gain, enhancing feed intake and decreasing the feed-meat ratio.
    Effects of Different Diet Combinations on the Growth Performance and Economic Benefits of Crossbred Beef Cattle in Winter

    The study aimed to study the effects of different feeding ways on the growth performance and economic benefits of beef cattle in winter. Eighteen crossbred beef cattle with body weight of about (201 ±21.46) kg , similar age and body size were randomly divided into 3 groups. The cattle in test group 1 was fed with 10 kg green alfalfa,2 kg mixed concentrate and corn stalk freely. The cattle in test group 2 were fed with 2 kg green alfalfa,8 kg green ryegrass,2 kg mixed concentrate and corn stalk freely. The cattle in test group 3 were fed with 5 kg green alfalfa, 5 kg green ryegrass, 2 kg mixed concentrate and corn stalk freely. The results showed that the total weight, daily gain per head,daily weight gain income and gross income in test group 1 were significantly different with that in test group 2 (P〈0.05). These indices in test group 3 had significant difference with test group 2 (P〈0.05)and no significant difference with test group 1(P〉0.05). Therefore the feeding method in test group 2 had better effects.
    Effects of Different Grazing Patterns on Vegetation Community Characteristics of Desert Steppe

    The vegetation community characteristics in desert steppe region in different grazing patterns were investigated. The results indicated that the height and density of Stipa tianschanica var. klemenzii were highest in all-day grazing region and its coverage was the highest in contrast region. The height of Cleistogenes mutica was the highest in contrast region, and its coverage and density were highest in the night-grazing region. The height and density of Leymus secalinus were higher in the day-grazing region and its coverage was the highest in contrast region. The overground biomass in night-grazing region was lower in July, August and September.

    41 lambs of Tibetan Sheep were purchased from Cuomei County of Shannan Prefecture in Tibet. The diarrhea of lambs that defecated watery putrid dark stools occurred after they arrived at Lhasa one day. Some lambs defecated the bloody stools or even died. The mixed infection of Escherichia coli and coccidian was confirmed by clinical diagnosis,postmortem and lab examination. The drug sensitivity results of E. coli isolated and cultured showed that E. coli was sensitive to gentamicin, amikacin, cephalosporinⅤand ceftazidime. Taking antisepsis and anti-inflammation, supplementing body fluid, preventing the electrolyte disorder and self acidosis and anti-shock as the treatment principles, a series of comprehensive measures were taken, and the diseased lambs were healed after 2 weeks.

    103 samples of cattle serum were collected from some areas of Xinjiang to detect the antibody of cattle mycoplasma by double-antigen sandwich ELISA method. The results showed that the mycoplasma detection results of 30 samples of serum were positive and the positive rate was 29.1%. This indicated that mycoplasma disease of cattle occurred in the investigated areas and the infection rate was higher. So some active measures should be taken to prevent and control the occurrence and prevalence of cattle mycoplasma.
