  • Analysis of OPN Gene Polymorphism and Its Association with Milk Production Trait in Guizhou Holstein Cows

    The aim of the present study was to assess the association between milk production trait and OPN gene polymorphism in Guizhou Holstein Cows. A total of 200 Guizhou Holstein Cows were used as experimental animals; a set of primers were designed to amplify and sequence the OPN gene by PCR-SSCP assay and DNA sequencing technique respectively, and subsequently the polymorphism of the OPN gene were analyzed; the association between milk production trait and OPN gene polymorphism were evaluated by using correlation analysis method. The results showed that there were two alleles for OPN gene including M and N in Guizhou Holstein Cows and their allele frequency was 0.62 and 0.38, respectively; the group polymorphism information content was 0.360, the heterozygosity was 0.689, and M allele was found as the dominant allele in the population; MM type individual had significantly or extremely significantly higher milk fat percentage and 305 days milk yield compared to NN type individual(P〈0.05 or P〈0.01), and had significantly higher 305 days milk yield compared to MN type individual(P〈0.05). The combined data suggest that MM genotype of OPN gene may serve as a molecular mark for selection of the traits of high milk fat percentage and milk yield in Guizhou Holstein Cows.
    Effects of Heavy Metal Lead and Mercury Exposure on Development and Antioxidant Enzyme Activity of Zebrafish Embryo

    In this study, the toxicological effects of the different concentrations of heavy metal ions(Pb^2+and Hg^2+) on zebrafish were evaluated. The embryonic mortality rate and hatching rate, deformity rate of juvenile fish, and the activity of superoxide dismutase(SOD) and glutathione S-transferase(GST-S) in embryonic cells were determined. The results showed that when zebrafish was exposed to different concentration of Pb^2+or Hg^2+for 8 to 92 hours, the embryonic mortality rate in treatment groups for both heavy metal ions was found significantly increased compared to the control groups(P〈0.05); the embryonic hatching rate exhibited a descending trend as the Pb^2+concentration increased, and the deformity rate of juvenile fish was increased when exposed to higher Pb^2+concentrations; no significant impacts of Hg^2+exposure on embryonic hatching rate were observed(P〉0.05), but 0.000 4 mg/L and 0.000 8 mg/L Hg^2+treatment groups had significantly higher deformity rate of juvenile fish compared to 0.000 2 mg/L Hg^2+treatment group(P〈0.05); the different types of malformation were observed in both Pb^2+and Hg^2+exposed juvenile zebrafish; as the extension of exposure duration, the SOD activity in embryonic cells for Pb^2+and Hg^2+treatment groups exhibited a gradually descending trend(P〈0.05) and a gradually ascending trend(P〈0.05), respectively; the GST-S activity was initially increased and then declined for both Pb^2+and Hg^2+treatment groups, and the greater changing range was observed in Hg^2+treatment group. In conclusion, the heavy metal ions of Pb^2+and Hg^2+had toxicological effects on zebrafish embryo, and the mortality rate and hatching rate of embryo and the deformity rate of juvenile fish were affected by regulating the activity of antioxidant enzymes(SOD and GST-S).
    Effects of Wenshenyijing Powder on Blood Physiological and Serum Biochemical Indexes in Rats

    The toxicity test is compulsory for the drugs to get the clinical approval. In recent years, the cases of poisoning and adverse reactions associated with traditional Chinese medicine are commonly reported, indicating that it is necessary to conduct toxicological evaluation on traditional Chinese medicine. In order to ensure the safety and reliability of the traditional Chinese medicine compound preparation Wenshenyijing Powder in clinical use and to objectively evaluate its safety, toxicity tests were designed and conducted in this study. The acute toxicity of Wenshenyijing Powder was assessed in accordance with the protocol recommended by ′ Veterinary Drug Acute Toxicity Test(LD50 Determination) Guidelines′. The long-term toxicity test was performed on rats which were assigned to four groups. Three experimental groups fed the basil diet containing 0.45, 0.15, 0.045 kg/kg of Wenshenyijing Powder respectively, and one control group fed the basil diet. The long-term toxicity test lasted for 30 days, and at the end of the test, the physiological and biochemical indexes of the male and female rats were determined. The results showed that Wenshenyijing Powder exhibited no toxic effect to rats.
    Isolation, Identification and Antimicrobial Resistance of Enterococcus faecalis from Swine in Henan Province

    The aim of the present study was to characterize the antimicrobial resistance of Enterococcus faecalis(E. faecalis)isolates from swine in Henan Province. A total of 300 batches of samples were collected from scaled pig farms; the isolation,culture, and purification of E. faecalis strains were performed by using conventional methods; the isolates were subsequently identified to species level by using automatic bacterial identification/antimicrobial susceptibility test system and PCR assay;micro-broth dilution method was used to evaluate the susceptibility of the isolates to antimicrobial agents. The results showed that a total of 154 strains were identified as E. faecalis and the overall isolation rate was 51.33%; high resistance rates were detected for cefoxitin, clindamycin, tiamulin, ceftiofur, sulfisoxazole, erythromycin and tilmicosin(all above 90%); isolates also exhibited resistance to doxycycline, florfenicol, penicillin and compound sulfamethoxazole, with resistance rates ranging from 60% to 90%.It is indicated that the antimicrobial resistance status of E. faecalis isolates from swine in Henan Province is serious.
    Development and Innovation of a Pushing Piece Dripping Pen Used in Blood Smear Preparation

    Currently,in the process of blood smears preparation, the blood samples are directly obtained from the open bottle. Due to the inconvenience of operation, mistakes are often made and the operation of pushing piece always fails. In order to overcome the shortcomings of existing technology, a pushing piece dripping pen used in blood smear preparation is developed in this study which consists of three parts including pen body, dripping head and the pushing plate components. The dripping head is set on the opening of the pen body with an inner diameter of 0.2-0.3 mm. The pushing plate components consist of a fixing sleeve and a pushing plate which is fixed in the fixing sleeve, and the fixing sleeve is set on the bottom of the pen body. Because the dripping of the blood samples is made by using dripping head, the dripping volume is easy to control and the operation is convenient. The structure of the pushing component being sleeve-connected to the pen body makes it easier to perform piece pushing and is convenient to be replaced. The application of the developed device provides assurance for the performance of teaching and experiment.
    Effect of Donkey Milk on Selected Physiological Indicators of Mice with Spleen Asthenia Syndrome

    The spleen asthenia syndrome model in mice was prepared by intraperitoneal injection of reserpine. The mice with spleen asthenia syndrome were given donkey milk by intragastric administration for fifteen consecutive days. The model control group and blank control group were also set in this study. After the last administration, the number of blood cells and the organ index of spleen and thymus were determined. The results showed that the number of blood cells and organ index of spleen and thymus of the model control group were significantly less and lower than those of the blank control group(P〈0.05); compared to the model control group, the donkey milk administration group had significantly more blood cells and significantly higher organ index of spleen and thymus(P〈0.05). The combined data suggest that donkey milk has a protective effect on the tissue cells of mice with spleen asthenia syndrome.

    In order to effectively solve the temperature controlling problems in staining process of paraffin section preparation,combing the advantages and disadvantages of the traditional staining jar, a temperature controllable staining jar is developed. An inner tank, a lid actively connected to the inner tank and an outer tank compose the staining jar. The inner tank is made with stainless steel; three devices are set around the outer tank, containing a chamber inside which is used to contain the inner tank,heater devices at bottom and internal sidewall, as well as a controlling module, a display module and a timing module outside which are connected to the heater devices by wires. During the staining process, the display module exhibits the temperature of the staining solution in inner tank; the temperature can be maintained in a range of 40-80 ℃ by regulating the controlling module; the staining duration can be preliminary set by using the timing module that can alert the experimenter at the end of the timing, which prevents the paraffin section from being stained too deeply or too lightly.

    In order to effectively solve the problems that blood smear slides and necessary reagents and items used in staining process encroach on the experimental platform space and wasted dyeing solution pollutes the environment, a staining scaffold used in blood smear preparation was developed. The staining scaffold is mainly composed of supporting frame, a number of partitions as well as the wasted liquid tank, staining platform and placing platform successively set from top to bottom of the staining scaffold.The slides smeared with blood sample are put evenly in the staining platforms and subsequently stained; after staining, the partitions are moved up to the placing platform and the staining and drying steps can be performed simultaneously; the remaining liquid produced in the process of smear drying flows to the wasted liquid tank and cannot drip directly. By using the developed staining scaffold, the preparation quality of the blood smear is improved, the duration for drying step is shortened, the centralized collection and treatment of the wasted liquid is beneficial for the biological safety, and the experimental platform space is saved.

    Effects of Different Interplanting Patterns on Productive Performance and Quality of Pennisetum purpureumcv. Guiminyin

    This study was aimed to screen the corresponding cultivation technique of forage with high yield and superior quality which was suitable to be promoted in Songtao Miao Nationality Autonomous County of Guizhou Province. The experiment was conducted using a randomized block design. The productive performance and quality of forage under different interplanting patterns with Pennisetum purpureum cv. Guiminyin and tall fescue were determined. The results showed that under the pattern of Pennisetum purpureum cv. Guiminyin interplanting two lines of tall fescue, the total fresh grass yield was up to 14666.53 kg/667 m^2,and the economic benefit was 5866.61 Yuan/667 m^2 which was 3926.48, 182.07, 601.31 Yuan/667 m^2 higher than that under the pattern of single planting tall fescue and Pennisetum purpureum cv. Guiminyin interplanting one line and three lines of tall fescue,respectively. In conclusion, the interplanting pattern of Pennisetum purpureum cv. Guiminyin + two lines of tall fescue not only greatly increases the forage yield in unit land area, but also meet the demand of animal husbandry development of Songtao region from March to September for green fodder and early spring forage feed. Furthermore, under this interplanting pattern, the forage exhibited superior nutritional quality and significant economic benefits are brought, indicating that the interplanting pattern is suitable to be promoted in this region.
    Analysis of Natural Grassland Vegetation Index in Hainan Prefecture of Qinghai Province Using TM Images and ARCGIS Software

    This study was aimed at assessing the vegetation index of natural grassland in Hainan prefecture of Qinghai province.The TM images band was extracted by using ARCGIS software to calculate ratio vegetation index, normalized difference vegetation index, difference vegetation index, soil-adjusted vegetation index, modified soil-adjusted vegetation index, and vegetation signal to soil noise. The results showed that the vegetation of alpine meadow was better than the other types of natural grassland. In the present study, the extracted vegetation information based on vegetation index is scarcely affected by soil background and this method has good extraction effect, indicating that vegetation index has the potential to be used to preliminary analyze the characteristics of natural grassland in Hainan prefecture of Qinghai Province.
    Characterization of Photosynthesis of Three Kinds of Leguminous Grasses in Different Months

    The present study aimed to characterize the photosynthesis of leguminous grasses in different months. Three kinds of leguminous grasses, including Melilotoides ruthenica, Lespedeza davurica, Medicago sativa, were selected as experimental materials. The environmental temperature(Ta) and the photosynthetically active radiation(PAR), stomatal conductance(Gs),transpiration rate(Tr), intercellular CO2 concentration(Ci) and net photosynthetic rate(Pn) of leaves of the three kinds of leguminous grasses were determined for three successive months, and the instantaneous water use efficiency(WUE) and light use efficiency(LUE) were calculated based on the related parameters mentioned above. The results showed that Melilotoides ruthenica was a low-photosynthesis, low-transpiration, and low-LUE typed plant, indicating that it was suitable to be planted in arid and low temperature regions; for Lespedeza davurica leaves, Pn was negatively correlated with Ci, LUE was very significantly positively correlated with Gs and significantly negatively correlated with WUE, and WUE was significantly positively correlated with Gs, indicating that they had stable ability in absorbing CO2; Medicago sativa was a high-photosynthesis, high-transpiration,and high-WUE typed plant whose leaves Pn was significantly positively correlated with Tr, indicating that it had significant CO2 fixation effect and was an ideal plant for alleviating greenhouse effect.

    Gene knockout is a kind of gene modification technology by which a specific gene with known sequences but without functional information can be inactivated or deleted via experiment designed at molecular level so as to speculate the biological function of the gene. Currently, this technology has been employed in many fields: in establishment of animal disease models, the application of gene knockout develops rapidly and it was widely used in human, mice, zebrafish, chicken, bovine, sheep, porcine,and other species; in functional genomics, the functional information of genes is found by establishment of gene knockout animal models; in pharmacology, gene knockout mice can be used to reveal the mechanism underlining the pathogenesis of diseases and to discover novel therapeutic targets; in immunology, the immune system diseases can be investigated at genetic level by gene knockout; in prevention and treatment of animal diseases, gene knockout is also wildly applied; in transgenic animal research, a lot of regulatory elements have been investigated for the external regulation of transgenic animals; gene knockout technology is also employed in research regarding replication, metabolism and pathogenesis of pathogenic microorganisms as well as growth,development and breeding of animals.
    Comparison of Pregnancy Rate of Two Embryo Transfer Methods in Sheep,Oviduct Transfer and Laparoscopic Uterine Horn Minimally Invasive Transfer

    [Objective]This study was aimed at evaluating and comparing the pregnancy rate of two embryo transfer methods in sheep, oviduct transfer and laparoscopic uterine horn minimally invasive transfer. [Methods]In the present study, Dorper Sheep were used as the donor and Mongolia Sheep served as the recipient; superovulation was induced in breeding Dorper Sheep by CIDR-FSH method, and estrus synchronization was induced in recipient by PRID +PMSG method; pregnancy rate of the above mentioned two methods was calculated and compared. [Results]50.6% of the recipients(n=178) treated by oviduct transfer were successfully pregnant and 73.4% of the recipients(n=109) received laparoscopic uterine horn minimally invasive transfer were successfully pregnant.[Conclusion]Oviduct transfer had significantly lower pregnancy rate compared to laparoscopic uterine horn minimally invasive transfer.
    Effects of Dietary Supplementation of Yeast Culture on Productive Performance and Economic Benefits of Beef Cattle

    This study was aimed at evaluating the effects of dietary supplementation of yeast culture on productive performance and economic benefits of beef cattle. A total of 150 Simmental crossbred cattle were randomly assigned to one of two groups such that control group(n=75) fed the TMR diet prepared in the experimental cattle farm and experimental group(n=75) fed the TMR diet supplemented with 0.5% of yeast culture. The experimental period lasted for 90 days. The results revealed that the daily gain and economic benefits were found increased by 9.68%(P〈0.05) and 9.09% in the experimental group compared to the control group respectively, and the ratio of feed to gain was found decreased by 1.79%(P〈0.05) in the experimental group compared to the control group. The results obtained in this study suggest that supplementing the basil diet with yeast culture improves the productive performance and economic benefits of beef cattle.
    Effects of Dietary Supplementation of Cinnamic Acid and Citric Acid Mixture on Productive Performance of Laying Hens

    The aim of the present study was to assess the effects of cinnamic acid and citric acid mixture supplementation on laying performance of hens during the peak period, and to evaluate the application value of the mixture as an alternative for antibiotics in livestock and poultry production. A total of 1000 24-week-old Hy-Line Brown Laying Hens were randomly assigned to 4 mixture-treated groups(30, 60, 120, 240 mg/kg) and one basal diet control group. The whole experiment lasted for 28 days during which the health status of the laying hens was observed and their productive performance was determined. The results showed that the diarrhea incidence and morbidity in 30 mg/kg group were significantly lower than those of the control group(P〈0.05), but the diarrhea incidence was increased as the higher dosages of the mixture was added. Compared to the control group, the significantly increased feed conversion rate(FCR) was observed in both 30 mg/kg and 60 mg/kg groups(P〈0.05). In conclusion, dietary supplementation of 30-60 mg/kg of cinnamic acid and citric acid mixture during the peak period is able to improve the productive performance of laying hens and to decrease the diarrhea incidence and morbidity.

    Xylo-oligosaccharide is a kind of functional oligosaccharide with higher additional value which is able to promote the proliferation of intestinal beneficial bacteria, suppress the growth of harmful bacteria, and regulate the intestinal microflora balance. Due to its smaller addition quantity and superior stability, xylo-oligosaccharide is widely used in the fields of feeding stuffs, food and medicine industry. The characteristics and application status in food industry and ruminate animal production of xylo-oligosaccharide is introduced, with emphasis on the research progress on its protective effect on calf diarrhea, in hoping to provide references for the scientific use of xylo-oligosaccharide in animal husbandry production.
    Estimation of Pollutants Production from Dairy Farms and Analysis of Corresponding Environmental Capacity in Erhai Lake Basin of Yunnan

    Based on the statistical data of dairy cows stocks in Erhai Lake Basin(Dali City and Eryuan County) from 2011 to 2015, the associated parameters were determined combined with the domestic research results to estimate the pollutants production from dairy cows breeding in Erhai Lake Basin of Yunnan, and the corresponding environmental capacity was also analyzed. The results showed that the average annual fecal and urinary emissions produced by dairy cows in Erhai Lake Basin of Yunnan from 2011 to 2015 were 595700 t and 268800 t respectively, and the average annual basic farmland load from fecal and urinary was 15.19 t/hm^2 and 6.85 t/hm^2 respectively; the average annual production of total nitrogen and total phosphorus were 4254.30 t and 492.66 t respectively, and the average annual basic farmland load from total nitrogen and total phosphorus were 108.47 kg/hm^2 and 12.56 kg/hm^2 respectively. The results obtained in this study provide references for the rational distribution and scientific division of dairy cow breeding industry in Erhai Lake Basin.

    With the implementation of the veterinary qualification examination system in China, the undergraduates majoring veterinary medicine are required to obtain the qualification of practicing veterinary medicine after graduation if they want to be engaged in the diagnosis and treatment of animal diseases. At the same time, with the increasing of the people′s living standard,pet ownership is rising rapidly and the market performance of pet clinic exhibited great development potential. This fact makes the universities and colleges pay more attention to the teaching of small animal medicine, but how to connect the small animal medicine specialty teaching with veterinary qualification examination is a problem that needs to be solved urgently. The teaching reform practice of small animal medicine direction in veterinary medicine specialty of Heilongjiang Bayi Agricultural University is explored and summarized, in hoping to provide references for the counterparts.

    Early realization of the Chinese dream is the main goal of the socialist modernization construction in China. The main purpose of this paper is to better integrate the Chinese dream into the ideological and political education of college students and guide the students to integrate their individual dream and the Chinese dream organically. Based on the summarization of the popularization approach of integration of the Chinese dream into the university′s ideological and political education, the joint point between the Chinese dream and the development characteristics of different groups of college students is focused from the following aspects: pioneer demonstration of CCP members, self-reliance of students from families with economic difficulties,academic truth pursuit of the students who take the postgraduate examination, practical ability of the students with innovative and entrepreneurial capability, so as to improve the diversity and effectiveness of realization approach of integration approach of the Chinese dream into the university′s ideological and political education.

    In recent years, the full-time master′ s cultivation programs in agriculture taken by Heilongjiang Bayi Agricultural University provide reserve and guarantee of talents for the agricultural modernization of the Heilongjiang reclamation area.However, there are still a lot of unsolved problems, such as the lack of clarity in education objectives and deviations in the directions taken by management.     Application of Task-driven Method in Animal Husbandry Microbiology Practice and Evaluation on Its Utilization Effect

    In order to improve the teaching effect of Animal Husbandry Microbiology practice, task-driven method was applied in practical teaching. A total of 62 students from 2 classes were randomly divided into four test groups. Task-driven method and conventional method were applied respectively. Students were scored based on the results of task completion and questionnaire.The results showed that the students in group 1 and group 3 who were taught with task-driven method had a much better grasp of practical contents compared to the students in group 2 and group 4 who were taught with conventional method, and the former had higher participation and enthusiasm for experiment content compared to the latter. In written examination of experimental course,there were 12 and 15 students in task-driven method taught groups who were able to completely master the knowledge they had learned respectively, accounting for 85.0% and 93.7% of the total number of students respectively; there were 5 and 6 students in conventional method taught groups who were able to completely master the knowledge they had learned respectively, accounting for 33.3% and 37.5% of the total number of students respectively, but they had lower participation and enthusiasm compared to the students in task-driven method taught groups. The results proved that the introduction of task-driven method in the Animal Husbandry Microbiology practice can obviously improve the teaching effect.

    Autophagy is a highly conserved and lysosomal-mediated process for the degradation of intracellular proteins and organelles to maintain the intracellular environment. Autophagy is involved in cellular physiological processes that affect the progression of some human diseases, including cancer, inflammation, and nervous system diseases. Meanwhile, it also acts as a cellular defense mechanism to prevent the host from infections caused by certain pathogenic bacteria and virus. At present, there are many ways to study autophagy: observing the morphological structure of autophagosome by transmission electron microscopy;estimating the autophagic activity by calculating the size and number of the tested autophagosome; determining the occurrence of autophagy in cells by testing the presence of autophagy protein LC3 in autophagosome membrane; detecting the autophagosome by monodansylcadaverine(MDC) staining method; determining the autophagic flux. The commonly used detection methods for autophagy are introduced, in hoping to provide references for the autophagy researchers.

    Pigs per mated sow per year (PSY) is a comprehensive index to evaluate the productive performance of sows and is of great significance for the efficient and sustainable development of pig farms. The facilities and operations of pig production should be pig-oriented and the precise management should be implemented, so as to make full use of pig potential and to achieve stable and high yield. The fine management techniques for increasing annual productivity of sows are discussed from the aspects of enhancing infrastructure construction in pig farms, introducing welfare concept into pig production, selecting breeding pigs with superior productive performance, and improving survival rate of weaned piglets.
    Investigation on Immune Status of Rabies in Dogs in Wenzhou

    In order to determine the seroprevalence of rabies virus among dogs in Wenzhou of Zhejiang Province, plasma samples of 351 dogs were tested forIgG antibodies against rabies virus by ELISA. The results revealed that rabies virus IgG antibodies were detected in 163(46.44%) of the 351 dogs; male dogs had slightly higher antibodies positive rate compared to female ones(P〉0.05), and the seroprevalence was extremely significantly higher in neutered dogs than intact dogs(P〈0.01); extremely significantly higher seroprevalence was observed in adult dogs compared to young(P〈0.01) and geriatric dogs(P〈0.01); the seroprevalence in household dogs was extremely significantly higher than dogs in training field(P〈0.01) and stray dogs(P〈0.01). The combined data suggest that the rabies virus prevails in dogs at a moderate level in Wenzhou, the management of dogs in training field and stray dogs should be enhanced, and the immunization coverage against rabies virus for dogs should be extended, so as to control the spread of rabies in this region.
